      目的 利用实时荧光定量PCR(real-time quantitative PCR, qPCR)法评价甘薯蛋白(sweet potato protein, SPP)对结直肠癌移植瘤荷瘤鼠肿瘤相关因子的调控作用。方法 12只雄性Babl/c裸鼠腹腔接种人结直肠癌LOVO细胞后经灌胃给予SPP干预21 d, 通过qPCR和免疫组化技术检测肿瘤组织中胰岛素样生长因子1受体(insulin like growth factor 1, IGF1R)、环氧酶2(cyclo-oxygen-ase 2, COX2)、血管内皮生长因子A(vascular endothelial growth factor A, VEGFA)、β链蛋白(β-catenin)的表达和分泌情况。结果 所有裸鼠均成瘤, SPP干预后, 肿瘤总重量及个数略减少, HE染色显示, 蛋白干预后肿瘤组织内小血管数量减少, 纤维组织增加。免疫组化检测显示, SPP干预后, IGF1R、COX2、VEGFA、β-catenin表达量均显著减少。qPCR检测显示, 与模型组相比,低剂量SPP干预后, VEGFA mRNA的相对表达量显著降低, 中、高剂量SPP干预后IGF1R、COX2、VEGFA、beta-catenin mRNA的相对表达量均显著降低。结论 SPP具有下调结直肠癌细胞移植瘤荷瘤鼠肿瘤中IGF1R、COX2、VEGFA、β-catenin表达和分泌的作用。

      Objective To evaluate the regulation of sweet potato protein (SPP) on tumor-associated factors in colorectal cancer xenograft nude mice by real-time quantitative PCR (qPCR). Methods After intraperitoneal inoculation of LOVO cells from human colorectal cancer in 12 male Babl/c nude mice, SPP was administered by perfusion for 21 d. The expression and secretion of insulin-like growth factor 1 receptor (IGF1R), cyclo-oxygen-ase 2 (COX2), vascular endothelial growth factor A (VEGFA) and β-catenin in tumor tissues were detected by qPCR and immunohistochemistry. Results All inoculated nude mice had tumors, the number and weight of tumors in the SPP intervention group were slightly decreased . HE staining showed that after the intervention of SPP, the number of small blood vessels was reduced, fibrous tissue was increased in the tumor tissue. Immunohistochemistry examination showed that after intervention, the expression levels of IGF1R, COX2, VEGFA and β-catenin were significantly decreased. QPCR showed that the mRNA relative expression of VEGFA was significantly decreased in the low-dose SPP group. In the medium-dose and high-dose SPP groups, the mRNA relative expressions of IGF1R, COX2, VEGFA and beta-catenin were all significantly decreased. Conclusion SPP has the effect of down-regulating the expression and secretion of IGF1R, COX2, VEGFA and β-catenin in the nude mice that transplanted colorectal cancer cells.
