      目的 采用响应面法优化确定有机双水相超声辅助萃取黄秋葵叶β-胡萝卜素的最佳条件并用大孔吸附树脂对其进行纯化。方法 采用乙醇-正丁醇超声辅助萃取黄秋葵叶β-胡萝卜素, 以β-胡萝卜素得率为考察指标, 采用响应面法研究乙醇质量分数、正丁醇质量分数、超声温度和超声时间对提取效果的影响。对超声辅助萃取所得的β-胡萝卜素粗提液用大孔吸附树脂进行纯化分离, 研究其静态、动态吸附曲线, 上样吸附及洗脱条件。结果 超声辅助萃取黄秋葵叶β-胡萝卜素的最佳条件为乙醇质量分数9%、正丁醇质量分数35%、超声时间3 min、超声温度40 ℃, 此条件下β-胡萝卜素的得率为9.12 mg/g。优选X-5型树脂并以乙醚为洗脱剂, β-胡萝卜素上样质量浓度为400 μg/mL, 上样吸附流速和洗脱流速为1 mL/min时纯化效果最好, 此条件下洗脱液中β-胡萝卜素的质量浓度最高为326.54 μg/mL。结论 本方法操作简易, 耗资低, 适合扩大生产。

      Objective To optimize and determine the optimal conditions of organic aqueous two-phase system combination with ultrasonic-assisted extraction of β-carotene from okra leaves by response surface method and purify it with macroporous adsorption resin. Methods β-carotene was extracted by ultrasonic-assisted extraction of ethanol-n-butanol from okra leaves. Taking the yield of β-carotene as an index, the effects of ethanol mass fraction, n-butanol mass fraction, ultrasound temperature and ultrasound time on the extraction were studied using response surface method. The β-carotene crude extract obtained by ultrasonic-assisted extraction was purified and separated with macroporous adsorption resin, and its static and dynamic adsorption curves, sample adsorption and elution conditions were studied. Results The optimal conditions for ultrasonic-assisted extraction of β-carotene from okra leaves were ethanol mass fraction of 9%, n-butanol mass fraction of 35%, ultrasound time of 3 min, and ultrasound temperature of 40 ℃. Under these conditions, the yield of β-carotene was 9.12 mg/g. The optimal purification condition was as follows: X-5 resin was used as the eluent, diethyl ether was used as the eluent, the mass concentration of β-carotene loading was 400 μg/mL, and the loading adsorption flow rate and elution flow rate were 1 mL/min, and the maximum mass concentration of β-carotene in the eluent was 326.54 μg/mL. Conclusion This method is easy to operate, low cost, and suitable for expanding production.
