      目的 分析3种不同固定相对皂苷异构体的分离效果。方法 3对皂苷异构体gypenoside L和gypenoside LI、damulin A和damulin B、20(S)-ginsenoside Rg3和20(R)-ginsenoside Rg3, 通过C18色谱柱、五氟苯基丙基色谱柱及双苯基色谱柱进行分离比较。以Inertsill ODS-SP色谱柱、Shim-pack Velox PFPP色谱柱和Shim-pack Velox Biphenyl色谱柱作为固定相, 用不同比例的流动相分离gypenoside L、gypenoside LI、damulin A、damulin B、20(S)-ginsenoside Rg3和20(R)-ginsenoside Rg3。结果 Shim-pack Velox PFPP色谱柱分别在29%和33%乙腈水溶液以1.0 mL/min等度洗脱, gypenoside L和gypenoside LI的分离度达到2.27, 20(S)-ginsenoside Rg3和20(R)-ginsenoside Rg3的分离度达到3.50, 分离效果最佳; Shim-pack Velox Biphenyl色谱柱在33%乙腈水溶液以0.5 mL/min等度洗脱, damulin A和damulin B的分离效果最佳, 分离度达到2.73。结论 五氟苯基丙基色谱柱因键合五氟苯基丙基, 对差向异构体具有更好的分离度, 双苯基色谱柱因键合二苯基, 对位置异构体有更好的分离度。

      Objective To analyze the separation effects of 3 different stationary phases on saponin isomer. Methods Three pairs of saponin isomers, gypenoside L and gypenoside LI, damulin A and damulin B, 20(S)-ginsenoside Rg3 and 20(R)-ginsenoside Rg3, were compared by C18 column, pentafluorophenylpropyl (PFPP) column and diphenyl column, respectively. Inertsill ODS-SP column, Shim-pack Velox PFPP column and Shim-pack Velox Diphenyl column were used as stationary phases, and gypenoside L, gypenoside LI, damulin A, damulin B, 20(S)-ginsenoside Rg3 and 20(R)-ginsenoside Rg3 were separated by different proportions of mobile phase. Results The effects of separation of gypenoside L and gypenoside LI, and 20(S)-ginsenoside Rg3 and 20(R)-ginsenoside Rg3 showed the best using Shim-pack Velox PFPP column. The resolution of gypenoside L and gypenoside LI was 2.27 eluted with 29% acetonitrile and the resolution of 20(S)-ginsenoside Rg3 and 20(R)-ginsenoside Rg3 was 3.50 eluted with 33% acetonitrile at flow rate of 1.0 mL/min; Damulin A and damulin B showed the best separation, with the resolution of 2.73, using Shim-pack Velox Diphenyl column eluted with 33% acetonitrile at the flow rate of 0.5 mL/min. Conclusion Pentafluorophenylpropyl column is used to separate the epimers due to the bonding of pentafluorophenylpropyl. Diphenyl column shows better resolution for positional isomers due to the bonding of diphenyl groups.
